In the anime, sins are created during a failed human transmutation. When they are initially made, they are formless and grotesque and must be fed a diet of incomplete stones to finish the transmutation.

To kill a sin in the anime, you must first purge them of their pstone material. Whether this means removing the stones in them , or making them use up the materal. This is done through the use of the Flamel's Cross transmutation circle. This tranmutation requires that you have a bit of the human the person was before.
In the Manga:
To kill a in in the anime, you have to retrieve the incomplete stone from them. Or you have to keep killing them until the stone runs out of juice. Then they become mortal and you can kill them. (I also theorize they have a weakness against their own sin's punishment in Hell, for instance Lust is burnt, and Greed is boiled. However, this could just be artistic symbolism, and not have anything to do with actual world mechanics)
